ELISA: Use at an assay dependent dilution. WB: Use at an assay dependent dilution. This peptide can be used with studies using ab38962. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance 

Calpain 3, also known as calpain 94 or muscle calpain, is an intracellular, calcium dependent cysteine protease. The calpains have papain like activity, thus the pain nomenclature. The calpain family members consist of a common small subunit, and a large variable subunit. Domains in the large subunit include the amino terminal domain I, the proteinase domain II, domain III, and the EF hand domain IV. Unlike M calpain which is ubiquitously expressed, Calpain 3 is a member of the tissue specific calpains. Unlike the other calpains, the endogenous calpain inhibitor, calpastatin, does not seem to inhibit Calpain 3. The half life of native Calpain 3 is also much shorter; about 10 minutes. Mutations in Calpain 3 have been linked to Limb Girdle Muscular Dystrophy, and native Calpain 3 is known to act as a meat tenderizer. The latent large subunit is 94 kDa, and the amino terminal truncation at activation yields an approximately 64 to 62 kDa form. A cascade of smaller forms can be seen with further activation.
