Description

Rabbit polyclonal to Calpastatin - Linker between domains 2C and 3A of the long isoform

Tested applications

WBmore details

Cross reactivity

Reacts with

Mouse, Rat, Human

Immunogen

Synthetic peptide to the linker between domains 2C and 3A in the long isoform of human Calpastatin.

(Peptide available as ab41382.)

Epitope

ab28254 recognises the linker between domains 2C and 3A in the Calpastatin long isoform.

Positive control

IL1 beta treated human colorectal adenosarcoma cells lysate.

General notes

ab28254 will also bind to the non-reduced protein, and thus has application for immunoprecipitation, immunohistochemistry, and ELISA. Diluted antibody can be reused several times if not contaminated; store at 4°C with 0.05% Sodium Azide as preservative.

Applications

Show applications key

Our Abpromise guarantee covers the use of ab28254 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application notes

WB: 1/1000 when using colorimetric substrates such as BCIP/NBT and 1/5000 for chemiluminescent substrates. Detects a band of approximately 82, 110 kDa (predicted molecular weight: 76 kDa). Higher concentrations of antibody may be needed for samples from more distantly related species Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Target

Function

Specific inhibition of calpain (calcium-dependent cysteine protease). Plays a key role in postmortem tenderization of meat and have been proposed to be involved in muscle protein degradation in living tissue.

Sequence similarities

Belongs to the protease inhibitor I27 (calpastatin) family.

Domain

Has four inhibitory domains.

Post-translational
modifications

The N-terminus is blocked.
