Abcam’s Interleukin-8 (IL-8) Canine ELISA Kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of dog IL-8 in serum, plasma and cell culture supernatants.
This assay employs an antibody specific for dog IL-8 coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-8 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-dog IL-8 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-8 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Function
IL-8 is a chemotactic factor that attracts neutrophils, basophils, and T-cells, but not monocytes. It is also involved in neutrophil activation. It is released from several cell types in response to an inflammatory stimulus. IL-8(6-77) has a 5-10-fold higher activity on neutrophil activation, IL-8(5-77) has increased activity on neutrophil activation and IL-8(7-77) has a higher affinity to receptors CXCR1 and CXCR2 as compared to IL-8(1-77), respectively. -
