Observed band size : 30 kDa (why is the actual band size different from the predicted?)
Additional bands at : 55 kDa (possible non-specific binding).
ab6619 at 1/2000 staining Carbonic Anhydrase I by Western Blot.
Lanes 1-3 are purified CA-I. The other lanes are 1ul human vitreous (1-3 ug protein). NDM: no diabetic patient, noDR: diabetic patient no diabetic retinopathy, QPDR: quiescent proliferative diabetic retinopathy, PDR: proliferative diabetic retinopathy. A HRP conjugated donkey anti-goat antibody was used as the secondary. The western blot shows that CA-I was only detected in vitreous from PDR and QPDR patients. (See Gao, et al. NATURE MEDICINE, 2007;13(2);181-188). Because the samples differ between patients and the severity of hemorrhage are also different so the intensity of the bands varies a lot. Another band was detected around 55 kDa and this was confirmed to be IgG.

Lanes 1-4 shows Carbonic Anhydrase I (positive control). Rat RBCs and human RBCs were loaded at 0.01ul. An HRP conjugated anti-goat antibody was used as the secondary.
Only 1 band at around 30kDa was detected using RBCs.
