Human Carcino Embryonic Antigen isolated from hepatic metastasis of colon adenocarcinoma.
Perchloric acid extract of hepatic metastasis of colon adenocarcinoma. Epithelial cells in colon, bronchial and alveolar epithelium, and alveolar macrophages in lung, polymorphonuclear leukocytes and macrophages in spleen, and the superficial epithelial layer and parietal cells in stomach. In liver, predominantly bilary canaliculi are labelled.

Ie: Use at an assay dependent dilution. (Crossed. Only the CEA and a CEA like protein precipitation arches appear when using 12.5 µL antibody per square. No reaction is seen with 2 µL normal human plasma.)
IHC-Fr: Use at an assay dependent dilution.
IHC-P: 1/200 - 1/400.Perform enzymatic antigen retrieval before commencing with IHC staining protocol.
ELISA: Use at an assay dependent dilution. (In double antibody sandwich ELISA, no reaction is observed with normal human plasma stripped of CEA.)
ICC/IF: Use a concentration of 1 µg/ml

Immunocytochemistry/ Immunofluorescence - Carcino Embryonic Antigen CEA antibody (ab15987)

ICC/IF image of ab15987 stained HepG2 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab15987, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
