A.Lot #GR50957-1 of ab19615 (cTnI antibody) did not work at all, even after contact with Abcam Tech Service. The replacement lot we received, #GR58712-1, worked but was weak compared to previous lots of this antibody that we've used in the past: #1078886, #978771, and #667212, which were all very strong. All lots tested in our lab were used under identical conditions, and it was noted that all were similar or near similar stock concentrations. We would have preferred a replacement vial from one of these stronger lots if it were possible, not from a "GR-" lot, which do not seem to work as well.

The most recent vial of antibody a19615 failed in IHC. Previous vials have worked well.

The older lots that you mentioned are unfortunately sold out but I suspect that just the single vial was defective. We have not received any other complaints about the current lot or the one that failed for you.

We did it on a 24 wells plate.

Methods:

1. Media removed, cells rinsed twice with PBS 2. 400uL of ice-cold methanol

added, incubated for 6 minutes 3. Methanol removed, cells rinsed 3x with PBS

4. 400uL of 3% BSA in PBS added to all wells and removed after several

minutes 5. 250uL of Troponin antibody (1:50) added to sample wells 6. 250uL

of 3% BSA in PBS added to negative control wells 7. Cells incubated for an

hour, room temperature in dark 8. Cells rinsed 3x with PBS and twice with 3%

BSA in PBS 9. 250uL of secondary antibody added 10. Cells incubated for an

hour, room temperature in dark 11. Cells rinsed 3x with PBS 12. 200uL of

Hoescht added to all wells, incubated for 10 minutes, room temperature in

dark 13. Cells rinsed 3x with PBS 14. View cells under fluorescence

microscope

Thank you for taking the time to contact us with these details. I am sorry to hear the customer has had difficulty obtaining satisfactory results from this antibody. The details you have kindly provided will enable us to investigate this case for you and this is also helpful in our records for monitoring of quality. Reviewing this case, I would like to offer some suggestions to help optimise the results from ab19615. I would also appreciate if you can confirm some further details: 1. Please provide the order number and date of purchase 2. Please confirm the antibody storage conditions 3. Are the cells allowed to dry after methanol removed and before rehydrating with the PBS? If the PBS is put on before the methanol has all evaporated, this can damage the cells. 4. I can recommend to try a different fixation. Try 4% PFA for 10 minutes only. You will need to include a different permeabilization step in this case, try 0.1% Triton for 10 minutes only. 5. As Cardiac Troponin I is a cytoplasmic protein, it is important to keep the cells permeabilised throughout the procedure. Try including 0.1% Tween 20 (a more gentle detergent that Triton) in the antibody dilution buffer, wash buffer and blocking buffer. 6. I can recommend to try incubating at 4oC overnight. Often provides more efficient and specific staining. 7. Please confirm if the secondary antibody working with other primary antibodies? Could you confirm if should detect the mouse IgG2b isotype? Please check and please provide the supplier and product code for the secondary antibody. In the event that a product is not functioning in the applications cited on the product data sheet (and the problem has been reported within 6 months of purchase), we will be pleased to provide a credit note, free of charge replacement or refund. One our customers wants to purchase antibodies to test in sELISA and human samples.

They want to detect cardiac troponins T and I.

1) anti-cardiac troponin T : ab45932 to coat on surface, then the protein ab47005 and then the FITC conjugated antibody ab105439 for detection.

2) anti-cardiac troponin I : ab27972, then Cardiac Troponin I protein ab50803 and ab56357 that will be conjugated with FITC using the Easylink kit.

Would it work for their sELISA experiments?

- ab105439 as detection antibody (directed against amino acids 64-82), www.abcam.com/ab105439 .

2) For the detection of Cardiac Troponin I :

- ab50803 as protein target, www.abcam.com/ab50803 ,

Thank you for your helpful reply. Regarding Cardiac Troponin I protein ref. ab9936, I see that it has been only tested for WB. Do you have information using it for ELISA? In other case, do you have a specific one for ELISA?  Regards,

The ab9936 product is a peptide which can be used to block antibodies ab7829, ab8290 and ab8291. This peptide can be used to block these antibodies irrespective of applications e.g. It is suitable to use with antibodies in Immunoprecipitation, western blot, Immunohistochemistry, ELSIA & also other blocking assays.
