    The Caspase 2 Colorimetric Assay Kit provides a simple and convenient means of assaying the activity of caspases that recoginse the sequence VDVAD.

    Caspase 2 Colorimetric assay is based on spectrophotometric detection of the chromophore p-nitroanilide (pNA) after cleavage from the labeled substrate VDVAD-pNA. The pNA light emission can be quantified using a spectrophotometer or a microtiter plate reader at 400- or 405-nm. Comparison of the absorbance of pNA from an apoptotic sample with an uninduced control allows determination of the fold increase in caspase 2 activity.

    Relevance

    Caspase 2 has a positive and a negative role in the regulation of the apoptotic process. Overexpression of the full length Caspase 2 (Nedd2/Ich1 ) mRNA in some cell types results in apoptosis, whereas overexpression of an alternative Caspase 2 splice variant suppresses apoptosis induced by serum withdrawal. Caspase 2 may be activated in vitro by Caspase 1, Caspase 3 and the neutral serine protease granzyme B which is stored in the specialized lytic granules of cytotoxic lymphocytes.
