  • Applications

Our Abpromise guarantee covers the use of ab51994 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

    Application Notes
    Functional Studies
  • Application notesFuncS: Caspase activity assay.
We recommend using 1 unit/assay for analyzing caspase activity.
One unit is the enzyme activity that cleaves 1 nmol of the caspase substrate WEHD-p-nitroanaline per hour at 37°C in a reaction solution containing 50 mM Hepes, pH 7.2, 50 mM NaCl, 0.1% Chaps, 10 mM EDTA, 5% Glycerol, and 10 mM DTT.

Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

  • FunctionInvolved in the activation cascade of caspases responsible for apoptosis execution. Cleaves caspase-1.
  • Tissue specificityWidely expressed, with highest levels in spleen and lung. Moderate expression in heart and liver, low expression in skeletal muscle, kidney and testis. Not found in the brain.
  • Sequence similaritiesBelongs to the peptidase C14A family.
    Contains 1 CARD domain.
  • Post-translational
    modifications
The two subunits are derived from the precursor sequence by an autocatalytic mechanism or by cleavage by Caspase-8.
