DESCRIPTION OF THE PROBLEM Does the casp9 antibody ab2013 detect your casp9 protein ab52203? If not can you recommend an antibody that will detect it so I can order the protein and ab together next week? I would like to perform a binding / inhibition experiment using these products if they are compatible. |
|||||||||
ANSWER: |
Thank you for your enquiry. Antibody ab2013 will recognize the recombinant CASP9 protein ab52203. Based on the units of ab52203 per vial received, you will receive about 62ug recombinant caspase 9 protein in a vial of ab52203. I hope this is helpful. Please contact me again if you have any further questions. |

Customer has found that ab2013 does not recognise the cleaved form of the protein following the supplier providing the information that it does. Offered a replacement vial of ab25758 as a replacement in return for an Abreview. |
|||||||||
ANSWER: |
Thank you for your enquiry. Further to our telephone conversation this morning I have updated our datasheet regarding the specificity of this antibody for the uncleaved form of Caspase 9. I look forward to reading your Abreview. I have arranged for a free of charge replacement vial of ab25758 to be dispatched immediately (order number: 135138). I hope that this does not affect your choice of Abcam in the future. |

Antibody MUST recognise the cleaved (and precursor) of Caspase 8 and 9 in WB |
|||||||||
ANSWER: |
Thank you for your email. I have performed a search of our catalogue and I can tell you that ab28142 and ab2013 are your best antibodies for the detection of cleaved and uncleaved caspase 8 and 9 by western blotting. The notes of each antibody are as follows: ab28142 detects ~55 and 57 kDa proteins, corresponding to the apparent molecular masses of the proform of caspase-8 alpha and beta on SDS-PAGE immunoblots. In apoptotic cells, this antibody also detects the processed forms (42/44 kDa, 31, 26, 18 and 14kDa). ab2013 detects both the activated (35 or 37kDa cleaved fragment depending on activator) and non-activated caspase-9 (45kDa). I would recommend that you have a look at the antibody's respective datasheets by clicking on the links below. I hope this information helps, please do not hesitate to contact us if you need any more advice or information. |

DOES THIS AB RECOGNIZE MOUSE CASPASE 9? CAN THIS AB DETECT ONLY THE 46 KDA PROTEIN? DOES IT CROSS REACT WITH THE ACTIVE CASPASE 9 (36 KDA)?
|
|||||||||
ANSWER: |
This antibody was made with a peptide located at amino acids 41-56 of human caspase-9. The peptide is located in the cleaved and activated fragment (35 or 37 kDa). Therefore, it should recognize the precursor and activated form although we have not tested the activated form. Our Western Blot tests show a strong, single band at 46kDa. It is human, mouse, and rat reactive. |
