IHC-P: Use a concentration of 20 µg/ml
IP: Use a concentration of 20 µg/ml
WB: Use a concentration of 1 - 4 µg/ml.Detects a band of approximately 30 kDa (predicted molecular weight: 38 kDa).Can be blocked with Cathepsin S peptide (ab54080). (One customer reports that overnight incubation with this antibody at room temperature gave the best staining in Western blot.)
IHC-Fr: Use a concentration of 20 µg/ml
Cathepsin family of proteases contains several diverse classes of enzymes. The cysteine protease class comprises cathepsin B, H, K, L, O, and S. The aspartyl protease class contains cathepsin D and E. Cathepsin G belongs to the serine protease class. Cathepsins are involved in various cellular events such as peptide biosynthesis, protein degradation, and apoptosis. Cathepsin S has been shown to be able to function as an elastase over a broad pH range in alveolar macrophages. Transcript variants utilizing alternative polyadenylation signals exist for this gene.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Cathepsin S antibody (ab18822)

IHC image of ab18822 staining in human tonsil formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol B. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab18822, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
