May act as a scaffolding protein within caveolar membranes. Interacts directly with G-protein alpha subunits and can functionally regulate their activity (By similarity). Involved in the costimulatory signal essential for T-cell receptor (TCR)-mediated T-cell activation. Its binding to DPP4 induces T-cell proliferation and NF-kappa-B activation in a T-cell receptor/CD3-dependent manner. Recruits CTNNB1 to caveolar membranes and may regulate CTNNB1-mediated signaling through the Wnt pathway.
Expressed in muscle and lung, less so in liver, brain and kidney.
Defects in CAV1 are the cause of congenital generalized lipodystrophy type 3 (CGL3) [MIM:612526]; also called Berardinelli-Seip congenital lipodystrophy type 3 (BSCL3). Congenital generalized lipodystrophies are autosomal recessive disorders characterized by a near absence of adipose tissue, extreme insulin resistance, hypertriglyceridemia, hepatic steatosis and early onset of diabetes.

The initiator methionine for isoform Beta is removed during or just after translation. The new N-terminal amino acid is then N-acetylated.

Western blot analysis of the extracts from NIH/3T3 cells untreated or treated with H2O2 using Caveolin 1 antibod (ab39541) and Caveolin 1(Phospho-Tyr14) antibody.
Lane 1: Using ab39541;
Lane 2: Using ab39541 preincubated with synthesized peptide;
Lane 3: Using Caveolin 1(Phospho-Tyr14) antibody;
Lane 4: Using Caveolin 1(Phospho-Tyr14) antibody preincubated with synthesized peptide.
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections)-Caveolin 1 antibody(ab39541)

Sections were stained using an automated system (DAKO Autostainer Plus), at room temperature: sections were rehydrated and antigen retrieved with the Dako 3 in 1 AR buffers EDTA pH 9.0. Slides were peroxidase blocked in 3% H2O2 in methanol for 10 mins. They were then blocked with Dako Protein block for 10 minutes (containing casein 0.25% in PBS) then incubated with primary antibody for 20 min and detected with Dako envision flex amplification kit for 30 minutes. Colorimetric detection was completed with Diaminobenzidine for 5 minutes. Slides were counterstained with Haematoxylin and coverslipped under DePeX. Please note that for manual staining we recommend to optimize the primary antibody concentration and incubation time (overnight incubation), and amplification may be required.
