Recommended dilutions IHC-P: Use at an assay dependent dilution. IHC-Fr: Use at an assay dependent dilution.
IHC: This antibody may be diluted to a titer of 1:25-1:50 in an ABC method. We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation conditions and antibody dilutions should be determined by the user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 1 mM EDTA buffer, pH 8.0 prior to immunostaining.
WB: 1/100 - 1/500. Predicted molecular weight: 34 kDa.
IP: 1/100 - 1/500.
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Cdk2 is a member of the Ser/Thr protein kinase family. It is highly similar to the gene products of S. cerevisiae cdc28, and S. pombe cdc2. It is a catalytic subunit of the cyclin-dependent protein kinase complex, whose activity is restricted to the G1-S phase, and is essential for cell cycle G1/S phase transition. This protein associates with and is regulated by the regulatory subunits of the complex including cyclin A or E, CDK inhibitor p21Cip1 (CDKN1A) and p27Kip1 (CDKN1B). Its activity is also regulated by protein phosphorylation. Two alternatively spliced variants and multiple transcription initiation sites of this gene have been reported.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections)-Cdk2 antibody [2B6](ab2363)
Ab2363 staining human placenta. Staining is localized to the nucleus. Left panel: with primary antibody at 4 ug/ml. Right panel: isotype control. Sections were stained using an automated system (Dako PT Link), at room temperature. Sections were rehydrated and antigen retrieved with the Dako 3-in-1 antigen retrieval buffer, EDTA pH 9.0. Slides were peroxidase blocked in 3% H2O2 in methanol for 10 minutes. They were then blocked with Dako Protein block for 10 minutes (containing casein 0.25% in PBS) then incubated with primary antibody for 20 minutes and detected with Dako Envision Flex amplification kit for 30 minutes. Colorimetric detection was completed with diaminobenzidine for 5 minutes. Slides were counterstained with Haematoxylin and coverslipped under DePeX. Please note that for manual staining we recommend to optimize the primary antibody concentration and incubation time (overnight incubation), and amplification may be required.
