Question 1

Tuesday 27-September-2011

I am very interested in antibodies that might discriminate between phospho-CDK1 and phospho-CDK2. ab76146 is described as detecting CDK2 phospho-Y15, but your site does not say whether it also detects CDK1 phosphorylated at this site (which is the same consensus sequence). Thanks.

ANSWER:

Thank you for your inquiry.

I have confirmed with the laboratory that we have not tested cross-reactivity with phospho-CDK1. Sequence analysis shows that the immunogen of ab76146 shares 100% identity with human CDK1. Thus, I would predict cross-reactivity, but it is not guaranteed without data.

Question 2

I am very interested in antibodies that might discriminate between phospho-CDK1 and phospho-CDK2. ab76146 is described as detecting CDK2 phospho-Y15, but your site does not say whether it also detects CDK1 phosphorylated at this site (which is the same consensus sequence). This is of concern since a recent paper as ifit was selective for phospho-CDK2 (Davies KD et al Cancer Biology and Therapy 12(9) Nov 2011 E.preprint). Canyou tell me your experience with this antibody and whether you believe it to be selective or not. If not, then I would also recommend you add this note on cross-reactivity to your website. Thanks.
