  • Relevanceab45275 provides 5mg of pure chicken erythrocyte native histone octamers. The octamers have been crystallized and the structure published to 1.90å resolution. The requirement for high yields of ultra pure proteins for X-ray crystallography studies and mass-spectrometry studies has stimulated a development programme to purify proteins from the nuclei of chicken erythrocytes under very gentle conditions. Thus native histone octamers can be produced without denaturation, with all the naturally post-translational modifications.

    • ab45275 separated using SDS-PAGE under non-reducing (lane 1) and reducing (lane 2) conditions. ab45275 contains the components of the histone octamer H3, H2B, H2A and H4.

    Thank you for contacting us.

The octamers are expected to be stable for a year, I am sorry we are unable to tell you how far they will be stable e.g. after 18 or 24 months - we unfortunaltey haven't done any stability test for this product.

I have finally received an answer from laboratory. They have mentioned that If the octamers are stored at -80 degrees in 20 % glycerol as supplied, they should OK after even a year. However when the samples are used, ...

I can now confirm that ab45275 can be dialyzed with leaving the octamers intakt by using 2M KCl, 0.2M K2HPO4, 0.2M KH2PO4, 2.5 mM benzamiding hydrochloride to dialyze into.

Histone core octamers only stay intac...

If the octamers are kept at -80C in 2M KCl, 0.2M K2HPO4, 0.2M KH2PO4, 20% glycerol, they may be thawed on standing at 4C for an hour or two. If they are dialysed into the buffer without the glycerol, they will be fine. Dialysis into 2M KCl with no buffer...

Histone core octamers are not intact except in high salt, in the case of the product that is 2M KCl, 0.2M K2HPO4, 0.2M KH2PO4. They break into histone dimers of H2A and H2B and histone tetramers of two copies of H3 and H4 if they are dialysed into 100mM ...

Please see also answer below. Nucleosome core particles can be constructed from the octamers, but assembly is to put tetramers on the DNA first and then the dimers. It is best to use defined sequences for the DNA. A protocol is in the reference: Asymm...

The octamers are intact and should crystallise as octamers to high resolution. However they should be treated as in the data and not allowed to be reduced in ionic strength from 2M KCl. 0.4 M equimolar potassium phosphate. They should be pretty well sto...
