Our Abpromise guarantee covers the use of ab14053 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Flow Cyt: Use 1µg for 106 cells.
ICC/IF: Use at an assay dependent dilution (PMID 19651890).
IHC-P: 1/100.
IHC-Fr: 1/100.
IHC-FoFr: 1/250.
WB: 1/500. Predicted molecular weight: 54 kDa.
Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.
Together with KRT19, helps to link the contractile apparatus to dystrophin at the costameres of striated muscle.
Observed in muscle fibers accumulating in the costameres of myoplasm at the sarcolemma membrane in structures that contain dystrophin and spectrin. Expressed in gingival mucosa and hard palate of the oral cavity.
Defects in KRT8 are a cause of cirrhosis (CIRRH) [MIM:215600].
Belongs to the intermediate filament family.
Phosphorylation on serine residues is enhanced during EGF stimulation and mitosis. Ser-74 phosphorylation plays an important role in keratin filament reorganization.
O-glycosylated at multiple sites; glycans consist of single N-acetylglucosamine residues.
Cytoplasm. Nucleus > nucleoplasm. Nucleus matrix.

Immunohistochemistry (Paraffin-embedded sections) - Cytokeratin 8 antibody (ab14053)

ab14053 at 1/100 dilution detecting Cytokeratin 8 from mouse ovary tissue sections by immunohistochemistry (paraffin-embedded sections). The antibody was incubated with the tissue for 16 hours and then detected using Alexa-Fluor® 647 goat anti-chicken antibody.

Immunohistochemistry (PFA perfusion fixed frozen sections) - Cytokeratin 8 antibody (ab14053)

ab14053 staining adrenal gland tissue sections by IHC-FoFr. Sections were PFA fixed and blocked with 0.5% TNB for 30 minutes at 25°C. The primary antibody was diluted 1/250 and incubated with the sample for 18 hours at 4°C. An Alexa Fluor® 546 conjugated goat anti-chicken antibody, diluted 1/500, was used as the secondary. The image (taken at 80x) demonstrates glandular cells in the adrenal gland.
