DNA polymerase delta is composed of 2 polypeptides present in equimolar ratio. The 125-kD subunit, POLD1, contains both the polymerase and the 3-prime-to-5-prime exonuclease activities, whereas the function of the 48-kD subunit is unknown. In addition to DNA replication, DNA polymerase delta is also involved in nucleotide excision repair of DNA, and PCNA, an accessory protein for DNA polymerase delta, is essential for both processes.
