Applications

Show applications key

Our Abpromise guarantee covers the use of ab3905 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Target

Function

Catalyzes the decarboxylation of L-3,4-dihydroxyphenylalanine (DOPA) to dopamine, L-5-hydroxytryptophan to serotonin and L-tryptophan to tryptamine.

Pathway

Catecholamine biosynthesis; dopamine biosynthesis; dopamine from L-tyrosine: step 2/2.

Involvement in disease

Defects in DDC are the cause of aromatic L-amino-acid decarboxylase deficiency (AADCD) [MIM:608643]. AADCD deficiency is an inborn error in neurotransmitter metabolism that leads to combined serotonin and catecholamine deficiency. It causes developmental and psychomotor delay, poor feeding, lethargy, ptosis, intermittent hypothermia, gastrointestinal disturbances. The onset is early in infancy and inheritance is autosomal recessive.

Sequence similarities

Belongs to the group II decarboxylase family.

ab3905 staining DOPA Decarboxylase in rat brain tissue sections by Immunohistochemistry (frozen sections). Tissue was paraformaldehyde fixed, then blocked with 5% serum for 1 hour at 25°C. Primary anitbody used at a 1/500 dilution and incubated for 16 hours at 25°C. The secondary was used at 1/200 goat anti-rabbit IgG conjugated to Alexa Fluor 488 (green).

Immunocytochemistry/ Immunofluorescence - DOPA Decarboxylase antibody (ab3905)

ab3905 staining DOPA Decarboxylase in mouse midbrain tissue sections by Immunohistochemistry (paraffin embedded sections). Tissue was fixed with paraformaldehyde and a heat mediated antigen retrieval step was performed using 10mM citrate buffer pH 6.0. Samples were then blocked with 1% serum for 30 minutes at 24°C followed by incubation with the primary antibody, at a 1/250 dilution, for 16 hour at 4°C. An Alexa Fluor® 488 conjugated goat anti-rabbit IgG (green) was used as secondary antibody at a 1/250 dilution.
