HeLa cells stained with ab51260 Dact1/Dapper homolog 1 antibody (red) at a dilution of 1/200.
.... cell extracts immunoprecipitated with ab51260 at 1/200 dilution (lane 2) or a negative control antibody (lane 1). Immunoprecipitates were blotted with ab51260 at 1/1000 dilution.
All lanes : Anti-Dact1 / Dapper homolog 1 antibody (ab51260) at 1/1000 dilution
Lane 1 : HEK293 cell lysate
Lane 2 : Molecular weight marker
Lane 3 : HEK293 transfected with Myc-Dact1 cell lysate
Predicted band size : 90 kDa
Observed band size : 90 kDa
Additional bands seen are thought to be degraded products of Dact1
IHC image of ab51260 staining in human hippocampus formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab51260, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
