WB: 1/250. Detects a band of approximately 115 kDa (predicted molecular weight: 82 kDa).
Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.
Negatively regulates the Nodal signaling pathway, possibly by promoting the lysosomal degradation of Nodal receptors.
Belongs to the dapper family.
The C-terminal PDZ-binding motif may mediate interaction with the PDZ domains of DSH (Dishevelled) family proteins.

Western blot - Dact2 antibody (ab42377)

All lanes : Anti-Dact2 antibody (ab42377) at 1 µg/ml
Lane 1 : Kidney (Mouse) Tissue Lysate
Lane 2 : Brain (Mouse) Tissue Lysate
Lane 3 : Kidney (Human) Tissue Lysate - adult normal tissue (ab30203)
Lane 4 : Brain (Human) Tissue Lysate - adult normal tissue (ab29466)
Lane 5 : HepG2 (Human hepatocellular liver carcinoma cell line) Whole Cell Lysate
Lane 6 : SK N SH (Human neuroblastoma) Whole Cell
Lysates/proteins at 20 µg per lane.
Secondary
Goat polyclonal to Rabbit IgG - H&L - Pre-Adsorbed (HRP) at 1/3000 dilution
Performed under reducing conditions.
Predicted band size : 82 kDa
Observed band size : 115 kDa (why is the actual band size different from the predicted?)
Additional bands at : 37 kDa. We are unsure as to the identity of these extra bands.
Exposure time : 1 minute
Western blot - Dact2 antibody (ab42377)

All lanes : Anti-Dact2 antibody (ab42377) at 1/250 dilution
Lane 1 : Pancreas (Mouse) Tissue Lysate (ab29363)
Lane 2 : NIH 3T3 (Mouse embryonic fibroblast cell line) Whole Cell Lysate (ab7179)
Lysates/proteins at 10 µg per lane.
Secondary
IRDye 680 Conjugated Goat Anti-Rabbit IgG (H+L) at 1/10000 dilution
Performed under reducing conditions.
Predicted band size : 82 kDa
Observed band size : 115 kDa (why is the actual band size different from the predicted?)
Additional bands at : 37 kDa,70 kDa. We are unsure as to the identity of these extra bands.
Dact2 has a predicted molecular weight of 82 kDa; however as this is a novel protein, it may contain uncharacterised modification sites that affect the migration of the protein. Abcam currently sell two antibodies to this target, ab42099 which detects a band at 100 kDa and ab42377 which detects a band at 115 kDa. Abcam welcomes customer feedback and would appreciate any comments regarding this product and the data presented above.
ab42377 has not yet been referenced specifically in any publications.
