Relevance

Dcp2 is necessary for the degradation of mRNAs, both in normal mRNA turnover and in nonsense-mediated mRNA decay. Dcp2 removes the 7-methyl guanine cap structure from mRNA molecules, yielding a 5'-phosphorylated mRNA fragment and 7m-GDP. It has higher activity towards mRNAs that lack a polyA tail. Has no activity towards a cap structure lacking a RNA moiety. Manganese is a cofactor that is required for highest activity. Dcp2 is found in a mRNA decay complex with LSM1, LSM3, LSM4, EXOSC2, EXOSC4, EXOSC10, PARN, XRN1, CNOT6, RENT1, RENT2 and RENT3B. Interacts with DCP1A, DPC1B, RENT1, RENT2 and RENT3B. The complex associates with polysomes.

ICC/IF image of ab28658 stained human HeLa cells. The cells were PFA fixed (10 min), permabilised in TBS-T (20 min) and incubated with the antibody (ab28658, 1µg/ml) for 1h at room temperature. 1%BSA / 10% normal goat serum / 0.3M glycine was used to quench autofluorescence and block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red). DAPI was used to stain the cell nuclei (blue).

ab28658 stainning Dcp2 in paraffin embedded human kidney. Kidney tissue sections (4 µm) were incubated with ab28658 (1/35 dilution) for 30 minutes at room temperature. Antigen retrieval was performed by heat induction in citrate buffer pH 6.

ab28658 was tested in a tissue microarray (TMA) containing a wide range of normal and cancer tissues as well as a cell microarray consisting of a range of commonly used, well characterised human cell lines. Further results for this antibody can be found at www.proteinatlas.org

Dcp2 contains a number of potential phosphorylation sites (Swiss Prot data) which may explain its migration at a higher molecular weight than predicted.
