I still have not heard anything back from the lab about the specificity of the 75kDa band. From what I have seen with the other Dicer antibodies in our catalogue and from other readings it does seem that the Dicer antibodies in general do recognize a band at 75kDa.\ I would be happy to send you another Dicer antibody from our catalogue if that would help you interpret your results. If you would like to test another antibody could you please let me know which one? |

I did another WB using your antibody. Again, I was looking for dicer in mouse kidneys. I used BSA 5% for blocking this time. The amount of protein per lane was the same as in the other WB before. Unfortunately, I don't get any band at 220kDa, only the one at 75kDa. I have attached the original .tiff-files with the integrated protein ladder for dicer. If you have any other suggestions, I am very willing to try them. Otherwise, I would ask you to suggest another antibody that we can use. |

ICC/IF image of ab13502 stained MCF7 cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab13502, 1µg/ml) overnight at +4ºC. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
