I am using this anti duck delta Fc antibody to detect IgY. In a reduced and denatured sample I see a light chain around 30kDa, a truncated heavy chain at between 40-50kDa but I do not see the full lenght heavy chain. Do you have any western blot data using this antibody on similar samples. |

ANSWER: |
I am sorry that we currently do not have in house datathat can be provided to the customer. However, from our records indeed the full length heavy chain can be seen. IT is possible the customer's samples are over-reduced, in particular if β-mercaptoethanol is being used. How are the samples reduced? |

Dears,We haveAnti-Rabies Virus antibody (ab67906), and our researcher want to evaluate this antibody for IHC application, I know that we will needRabbit polyclonal Secondary Antibody to Duck IgY - ΔFc (HRP) (ab31140) as secondary antibody... What else shall be need to conduct the experiment from Abcam? Urgent please. Thanks ! |

Thank you very much for your inquiry. Please find a list of links to reagents etc. that are needed for IHC. I also included a link to our extensive protocols page where comprehensive protocols for IHC and many more tips and tricks can be found: buffers and reagents: http://www.abcam.com/index.html?pageconfig=resource&rid=12854 mounting media: http://www.abcam.com/index.html?pageconfig=resource&rid=12862 substrate: http://www.abcam.com/index.html?pageconfig=resource&rid=12886 PAP pen: Click here (or use the following: http://www.abcam.com/index.html?datasheet=2601). Protocols: http://www.abcam.com/index.html?pageconfig=popular_protocols I would like to make you aware that ab67906 is not tested and guaranteed for IHC. The same is true for ab31140. I therefore suggest to use ab1002 (Anti-Rabies Virus antibody) which is tested and guaranteed for IHC-P and IHC-Fr. Click here (or use the following: http://www.abcam.com/index.html?datasheet=1002). A suitable secondary would be ab97040 (Goat polyclonal Secondary Antibody to Mouse IgG - H&L (HRP), pre-adsorbed). With this combination of antibodies I suggest to use goat serum for blocking in your IHC experiment: ab7481 (Goat Serum) Click here (or use the following: http://www.abcam.com/index.html?datasheet=7481). The blocking serum is chosen according to the host animal of the secondary, so if the duck antibody is used with the secondary anti duck form rabbit, I recommend to use rabbit serum: ab7487 (rabbit serum, sterile) Click here (or use the following: http://www.abcam.com/index.html?datasheet=7487). I hope you will find this information helpful. Please do not hesitate to contact me again with any further questions. |
