E2F8 (E2F transcription factor 8) is an inhibitor of E2F-dependent transcription. It binds DNA through the E2 recognition site, 5'-TTTC[CG]CGC-3' and regulates a subset of E2F-dependent genes whose products are required for normal cell cycle progession. It modulates the activity of other E2F proteins, such as E2F1, probably by competing or displacing activating E2Fs from promoter binding. E2F8 also acts as a guanine nucleotide exchange factor (GEF) for the alpha subunit of heterotrimeric G proteins. The molecular mechanism and biological significance of this effect remain to be determined.
