Applications

Show applications key

Our Abpromise guarantee covers the use of ab52032 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application notes

Peptide ELISA: Antibody detection limit dilution 1:64,000.
WB: Use at a concentration of 1.0 - 3.0 µg/ml. Detects a band of approximately 26 kDa (predicted molecular weight: 24 kDa). A minor band of unknown identity was also consistently observed at 75kDa. This band was successfully blocked by incubation with the immunising peptide.

Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Target

Relevance

EBAG9 is a type III transmembrane protein that is predominantly located in the Golgi. It induces apoptosis of natural killer (NK)/T cells and plays a role in immune invasion, possibly in relation to Epstein-Bar virus infection. EBAG9 is abundantly expressed in prostate cancer cells when compared to normal epithelial cells, and immunodetection of EBAG9 expression can be a negative prognostic indicator for patients with prostate cancer.

Cellular localization

Golgi apparatus; Golgi apparatus membrane; single pass type III membrane protein.

All lanes : Anti-EBAG9 antibody (ab52032) at 0.2 µg/ml

Lane 1 : A431 lysate
Lane 2 : A431 lysate with Immunising peptide

Lysates/proteins at 35 µg per lane.

Predicted band size : 24 kDa
Observed band size : 26 kDa (why is the actual band size different from the predicted?)
Additional bands at : 75 kDa. We are unsure as to the identity of these extra bands.

The band at 75kDa was successfully blocked by incubation with the immunising peptide.

Primary incubation was 1 hour. Detected by chemiluminescence.
