I purchased ab11424 a Rabbit Polyclonal to EDG1, and used it in WB and mouse which seems to be guaranteed on the datasheet. I had very weak bands and had better results with a similar ab from another company. |

Thank you for your inquiry. I heard back from the testing laboratory regarding the fixation/permeabilization protocol for ab11424. Aspirate culture medium and add warmed Fixation Solution (4% Formaldehyde, 37C) and incubate 15 minutes at room temperature. Aspirate Fixation Solution and wash with Dulbecco’s PBS. Aspirate solution. Add Permeabilization Solution (Dulbecco’s PBS with 0.1% Trition X100) and incubate 15 minutes at room temperature. Aspirate solution and proceed to blocking/antibody steps. This enquiry is in regard to EDG1 antibodies (both ab11424 and ab13126). Ultimately we would like to use an EDG1 antibody to stain formalin-fixed paraffin embedded tissue sections of rhesus macaques (monkeys). The product sheet for ab11424 states that the antibody can be used for immunofluorescence, but does this include immunofluorescent detection on tissue sections (fixed, frozen or paraffin embedded)? Ab13126 appears to work in formalin-fixed paraffin-embedded sections, but has not been tested in any other species except humans, is this antibody expected to cross-react with a broad range of species like the ab11424 antibody? Also, could either of these antibodies be used to stain unfixed cells for flow cytometric analysis?

Thank you for your email. When ab11424 was tested in IF, transfected CCL39 cells were used. So to our knowledge it has not yet been tested in IHC. Ab13126 has only been tested for cross-reactivity with human and we don't have any information about how it will work with other species.
