    ab100679 EGF (Epidermal growth factor) Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse EGF in serum, plasma and cell culture supernatants. This assay employs an antibody specific for Mouse EGF coated on a 96-well plate. Standards and samples are pipetted into the wells and EGF present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse EGF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of EGF bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

  • RelevanceEpidermal growth factor (EGF) has a profound effect on the differentiation of specific cells in vivo and is a potent mitogenic factor for a variety of cultured cells. The EGF precursor is believed to exist as a membrane bound molecule which is proteolytically cleaved to generate the 53 amino acid peptide hormone that stimulates cells to divide. EGF exerts its actions by binding to the EGFR, a 170 kDa protein. Epidermal growth factor (EGF) is a key growth factor regulating cell survival. Through its binding to cell surface receptors, EGF activates an extensive network of signal transduction pathways that include activation of the PI3K/AKT, RAS/ERK and JAK/STAT pathways. Because of its key role in driving the proliferation of cells, EGFR is a target of several anti-cancer drugs currently in development.
