I think this confusion is my mistake and sorry for

that.

 

Our customer labelled it in Korean so I

transferred it but, it's not right one.

 

before means diluted in

serum before aliquot and store -20C

 

and

 

this time means stored -20C

antibody  and diluted in serum right before using it.

 

 

The weaker signal was 'dilute in serum after

thaw'(=this time).

Thank you, this makes sense.

I think the addition of serum before freezing the aliquots of the antibody may be helping prevent loss of the antibody to the plastic tubes it is stored in. Typically, proteins will stick to plastic via hydrophobic interactions. The proteins in the serum will coat those surfaces, and so there is less surface for the antibody to stick to. More antibody is recovered from the tube. That is one explanation.

Another possibility is that antibodies that are stored with other proteins (for instance, bovine serum albumin or serum) tend to be more stable because they aggregate less readily with themselves during the thaw process than they would without the other proteins.

Antibody ab14016 is delivered in a formulation that does not contain any other proteins, just PBS. So your customer may be correct to add serum before freezing. However, we have not done this test, adding serum and then freezing and thawing, with this antibody. If there is a stabilizing effect of serum or other proteins, I do not think it is unique to ab14016.

This antibody has not been tested in IHC. However, from the SwissProt database, I see that the protein should be present in kidney or liver of adult mammals and liver of fetal or neonatal mammals.
