Specificity

Stains preferentially large vessels of the liver, lung capillaries, gall bladder and skin. Generally reactive with melanoma, including metastasizing melanomas and in carcinomas. Cross reactivity with nervous tissue has been reported.

Relevance

The endothelium is the layer of thin specialized epithelium, comprising a simple squamous layer of cells that line the interior surface of blood vessels, forming an interface between circulating blood in the lumen and the rest of the vessel wall (simple squamous epithelium). Endothelial cells line the entire circulatory system, from the heart to the smallest capillary. Endothelial cells are involved in many aspects of vascular biology, such as vasoconstriction and vasodilation, and hence the control of blood pressure; blood clotting (thrombosis & fibrinolysis); atherosclerosis; formation of new blood vessels (angiogenesis) and inflammation and swelling (oedema). Endothelial cells also control the passage of materials, and the transit of white blood cells, into and out of the bloodstream.

ab58821 staining mouse Endothelial cells derived from heart tissue by ICC/IF (Immunocytochemistry/immunofluorescence). Heart tissue was disassociated by the collagenase/trypsin method and the isolated cells were cultured on a chamber slide with DMEM/FCS medium. Cells were fixed with ice cold acetone and blocked with 5% BSA for 1 hour at 4°C. Samples were incubated with primary antibody 1/100 in PBS for 12 hours at 4°C. An Alexa Fluor® 555-conjugated Goat polyclonal to mouse IgG, dilution 1/500, was used as secondary antibody. Nuclei were counterstained by DAPI.
