What is the immunogen sequence for this antibody and do you know if it will react with mouse? |
|||||||||
ANSWER: |
Thank you for contacting Abcam. The lab has confirmed that as the immunogen for ab9774 was a stromal preparation of rat lymph nodes, no sequence is available. However, they did confirm that they could not demonstrate cross-reactivity with either human or mouse. Unfortunately the only endothelial cell antibody we have in our catalogue that reacts with mouse is ab58821 but it is unsuitable for IHC-P. There is a chance ab15605 may work but it has not been tested in mouse. As ab15605 has not been tested in mouse, I could offer you our testing discount program for this antibody. Please see the link below for more information: http://www.abcam.com/index.htmlpageconfig=resource&rid=11998&viapagetrap=collaborationdiscount If there is anything else I can help you with, please let me know. |

What is the immunogen sequence for this antibody and do you know if it will react with mouse? |
|||||||||
ANSWER: |
Thank you for calling Abcam yesterday. I have been in contact with the lab about ab9774 to see whether there is a possibility that it would recognize mouse tissue. Unfortunately the exact epitope site has not been mapped and so I am unsure as to whether this antibody will react with mouse tissue or not. There is a chance that it will work but we cannot guarantee it. As I cannot guarantee that the antibody will work for you, you could take advantage of our testing discount program, please see the details below: Testing Discount As ab9774 has not been tested in mouse tissue before,I can offer a discount off a future purchase if you buy ab9774 now, test it in mouse tissue and submit feedback to us in the form of an Abreview. It doesn’t matter whether the Abreview is positive or negative, we would just really like to receive your feedback. The discount would be to the value of 1 free PRIMARY ANTIBODY. If you are interested in this offer, please follow these steps: 1. Reply to this e-mail to let me know that you would like to proceed and test ab9774 in mouse tissueI will then send a discount code. This code must be issued before purchasing ab9774 so please wait for my reply before ordering. 2. Purchase ab9774 either by phone, fax, or online (www.abcam.com). 3. Test it in mouse tissue. 4. Let us know the results, positive or negative, using our Abreview system (this will take about 10 minutes and images are great if you have them!). To find out how to submit an Abreview, please visit: http://www.abcam.com/abreviews. 5. After the review is submitted to us, the discount code becomes active. Simply place your new order by phone, fax, or on the web and mention the discount code. The discount can be redeemed for any PRIMARY ANTIBODY ordered and the discount code is valid for 4 months after issue. We are always pleased to obtain feedback about our products and any information is greatly appreciated! Even if ab9774 turns out to be unsuitable for mouse tissue, you will still receive the discount on your next purchase after your Abreview has been submitted. Please let me know if you have any questions about this offer and I would be happy to help you further. The Terms and Conditions of this offer can be found at: www.abcam.com/collaborationdiscount. |

IHC image of Endothelial Cell staining in Rat brain FFPE section, performed on a BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab9774, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX
ab9774 staining cells of rat liver sections by IHC-Fr. Cells were acetone fixed and blocked with 1% BSA for 5 minutes at 25°C prior to incubating with ab9774 diluted 1/25 for 30 minutes at 25°C. An Alexa Fluor® 488 conjugated goat anti-mouse antibody was used as the secondary.

ab9774 staining endothelial cells in Rat lung tissue by Immunohistochemistry (Frozen sections). The sections were fixed in acetone prior to blocking with 100% SuperBlock Blocking Buffer for 20 minutess at 23ºC. The primary antibody was diluted 1/100 and incubated with the sample for 12 hours at 4ºC. A Biotin-conjugated Rabbit anti-Mouse polyclonal was used as the secondary antibody, diluted 1/200.
