Specificity
|
This antibody recognizes both the engrailed and invected gene products of Drosophila. The antibody is useful for examining the patterning of segmentation from the cellular blastoderm stage onward. Staining is also seen in a large number of CNS neurons including most of the median neuroblast progeny as well as a small number of PNS neurons.

Epitope has been localized to residues 38-58 of the homeodomain.

Recommended dilutions IHC-F: 1/200. ab12454 can be used with embryos fixed for 1 hour in phosphate-buffered 4% paraformaldehyde (pH 7.0) (Rowan et al.). Antigen retrieval is not normally necessary.
The antibody will recognize denatured proteins and is also valuable for expression cloning.
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
|
In Drosophila, the Engrailed gene plays an important role during development in segmentation, where it is required for the formation of posterior compartments. Expression studies have suggested that Engrailed may also function early in organizing the preblastoderm and later in neurogenesis. Engrailed encodes a homeodomain-containing protein that can function as a transcription factor. Invected is homologous to Engrailed and the gene lies adjacent to that for Engrailed. The function of Invected is unclear.

ab12454 used in immunohistochemistry at 1/200 on Drosophila embryos. The protocol used is available in the Neuroscience Abwire in the protocols section. Briefly, embryos were fixed with PBS, formaldehyde and n-heptane. Embryos were then dechorionated with 100% bleach and rehydrated with PBT (PBS with 0.3% Tritone added) before antibody staining.Visualisation was with DAB.
