Immunocytochemistry/ Immunofluorescence - Epithelial Antigen antibody [Ber-EP4], prediluted (ab82752)
Our Abpromise guarantee covers the use of ab82752 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
IHC-P: 1/1Perform heat mediated antigen retrieval before commencing with IHC staining protocol.
IHC-Fr: 1/1(Fix with acetone.)
ICC/IF: Use a concentration of 5 µg/ml
Epithelial antigen is a membrane bound, glycosylated phosphoprotein. The protein is anchored to the apical surface of many epithelia by a transmembrane domain, with the degree of glycosylation varying with cell type. It may may play a role in adhesive functions and in cell to cell interactions, metastasis and signalling and provide a protective layer on epithelial surfaces. It is capable of direct or indirect interaction with actin cytoskeleton.
Apical cell membrane; Single pass type I membrane protein. Note: Exclusively located in the apical domain of the plasma membrane of highly polarized epithelial cells. After endocytosis, internalized and recycled to the cell membrane. Located to microvilli and to the tips of long filopodial protusions.

ICC/IF image of ab82752 stained MCF7 cells. The cells were 4% PFA fixed (10mins) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab82752, 1ml) overnight at +4°C. The secondary antibody (green) was ab96879 Dylight 488 goat anti-mouse IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
