    ab100510 Human ErbB 2 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human ErbB 2 in serum, plasma and cell culture supernatants.

     

    This assay employs an antibody specific for Human ErbB 2 coated on a 96-well plate. Standards and samples are pipetted into the wells and ErbB 2 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human ErbB 2 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of ErbB 2 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

  • RelevanceErbB 2 is a receptor tyrosine kinase of the ErbB 2 family. It is closely related instructure to the epidermal growth factor receptor. ErbB 2 oncoprotein is detectable in a proportion of breast and other adenocarconomas, as well as transitional cell carcinomas. In the case of breast cancer, expression determined by immunohistochemistry has been shown to be associated with poor prognosis.

      The capture antibody is monoclonal and from mouse, while the detection antibody is polyclonal and from goat. Both antibodies detect the extracellular domain of ErbB2.
