I would like to order the peptide for the epitope of : Mouse monoclonal
[3B5] to ErbB 2, (IgG1) Synthetic peptide: TAENPEYLGLDVPV, corresponding
to C terminal amino acids 1242 - 1255 of Human c-ErbB2 (Ab16901). Is
ErbB 2 peptide (ab38955) the correct one , or do you have the one for
amino acids 1242 - 1255 of Human c-ErbB2? (ab38955: Synthetic peptide
derived from within residues 1200 to the C-terminus of Human ErbB 2.This
peptide was used as an immunogen for ab38956 - ErbB 2 antibody.)

As discussed previously, the band at ˜170 kDa may well be the specific band for ErbB 2 as the protein goes through post translational modification to increase its weight. In order to reduce the non-specific staining observed the customer could try as previously discussed:


1. to try with a different blocking agent (such as 5%BSA) we would typically recommend incubating with the blocking buffer for 1 hour at 4 degrees. A mild detergent such as Tween 20 (0.2%) in the dilution buffer of the primary antibody may also be beneficial.


2. lowering the concentration of the antibody, (0.1, 0.5 and 1 ug/mL) may improve the results (with incubation at 4 degrees overnight and 1 hour at room temperature if possible). It would also be useful to carry out a no-primary control to see if any of the non-specificity seen is due to the secondary antibody.

If these suggestions have been tried by your customer and they have not seen any improvement the I would be happy to offer a replacement antibody of a different lot, a different antibody against the same target (such as http://www.abcam.com/ErbB-2-antibody-ab2428.htmlwhich is a rabbit polyclonal which has been used with very positive results by many of our customers), or a credit note.

Thank you for sending us the detail information. It was very helpful. Our customer wanted to ask one additional quesiton. They applied this antibody to MCF7 (human breast cancer cell). They did not  have any good luck with this experiment. Could you please inform us any published paper where ab16901 was successfully used? If you cannot findany, could you recommand any good protocol he can use? FYI, he is treating ab16901 to repress tumeric activity in MCF7 cell. Thank you very much!

Dear Technical team, The customer purchased ab16901 and got several questions. 1. Is ab16901 specific for breast cancer ErbB2? Did you test this antibody against any other cnacer/tissue ErbB2 such as pancreatic cancer, bladder cancer? 2. Could you recommand an ErbB2 antibody that is specific for Human pancreatic cancer ErbB2? Thank you in advance for your help.

ab16901 is not specific to breast cancer but to ErbB2 protein that is expressed in a variety of tumor tissues including primary breast tumors and tumors from small bowel, esophagus, kidney and mouth.

As shown on NCBI (http://www.ncbi.nlm.nih.gov/UniGene/ESTProfileViewer.cgi?uglist=Hs.446352) the expression of ErbB2 in pancreatic tumor exists but it is not as high as in breast or colorectal tumors.
