Recommended dilutions Flow Cyt: Use 10µl for 105 cells. IHC-P: Use at a concentration of 10 µg/ml. This antigen is known to be sensitive to fixation: Trypsin or Proteinase K (6 min, RT) enzyme pretreatment is recommended for tissues fixed for less than 24 hours. Citrate buffer pH6.0 is recommended for tissues fixed for more than 24 hours. IHC-Fr: Use at a concentration of 10 µg/ml. IHC (PFA fixed): Use at a concentration of 10 µg/ml. IHC-FoFr: Use at a concentration of 10 µg/ml. IHC-R: Use at a concentration of 10 µg/ml. IP: Use at an assay dependent dilution. RIA: Use at an assay dependent dilution. WB: Use at a concentration of 10 µg/ml. Use under non reducing conditions. Detects a band of approximately 160 kDa (predicted molecular weight of precursor protein: 102 kDa; protein is heavily glycosylated). Block in 5% milk for 1 hour.
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

F4/80 antigen is a 160 kDa cell surface glycoprotein that is a member of the EGF TM7 family of proteins which shares 68% overall amino acid identity with human EGF module containing mucin like hormone receptor 1 (EMR1). Expression of F4/80 is heterogeneous and is reported to vary during macrophage maturation and activation. The F4/80 antigen is expressed on a wide range of mature tissue macrophages including Kupffer cells, Langerhans, microglia, macrophages located in the gut lamina propria, peritoneal cavity, lung, thymus, bone marrow stroma and macrophages in the red pulp of the spleen. F4/80 expression has also been reported on a subpopulation of dendritic cells but is absent from macrophages located in T cell areas of the spleen and lymph node. The ligands and biological functions of the F4/80 antigen have not yet been determined but recent studies suggest a role for F4/80 in the generation of efferent CD8+ve regulatory T cells.

Ab6640 positively staining (1/100) formaldehyde fixed mouse spleen tissue. Enzymatic antigen retrieval was carried out using "Digest-All" incubated for 5 minutes at 37oC.Detection was achieved using DAB. Ab6640 at a 1/8000 dilution positively staining formaldehyde fixed mouse spleen tissue. Enzymatic antigen retrieval was performed and the antibody was incubated for 10 minutes. Ab6733, a rabbit polyclonal to rat IgG H&L (biotin), was used as the secondary antibody at a dilution of 1/400. Immunohistochemistry (PFA perfusion fixed frozen sections) - F4/80 antibody [CI:A3-1] (ab6640)
ab6640 staining mouse brain tissue sections by IHC-FoFr. Sections were PFA fixed and blocked with 0.5% TNB for 30 minutes at 25°C. The primary antibody was diluted 1/10 and incubated with the sample for 18 hours at 4°C. A biotinylated goat anti-rat antibody, diluted 1/500, was used as the secondary.
