I am trying to source an antibody that works against pig FADD. We have already tried the Stressgen antibody raised against the peptide from the human sequence KIDSIEDRYPPNLTER, and it did not work on pig cell lysates (contrary to the data sheet) but worked on human cell lysates. Upon closer examination, I found that the pig sequence is closer to the mouse peptide in this region and there are quite a number of differece between the pig and human sequences. The abcam antibody ab24533 is raised against a peptide that includes this region, but your website only gives a range for the peptide of aa 100-150 and no precise sequence info. Does your peptide include the region ICDNVGKDWRRLARQLKVSD, which is immediately N terminal to the KIDSIEDRYPPNLTER sequence and a perfect match with the pig (Sus scrofa) sequence? I am basically wanting to know whether or not there is a chance that the abcam antibody will work on pig cells. Also, the dog and pig sequences are almost identical. Thank you |

What residues does it correspond to on FADD? The product sheet states that it surrounds aa 128 of human FADD. I'm not sure exactly which residues it corresponds to. |

ICC/IF image of ab24533 stained HepG2 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab24533, 5µg/ml) overnight at +4ºC. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
IHC image of ab24533 staining in human normal lymph node formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab24533, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
