Abcam’s Fibroblast growth factor (FGF2) basic mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of mouse FGF basic in serum, plasma and cell culture supernatants.

This assay employs an antibody specific for mouse FGF basic coated on a 96-well plate. Standards and samples are pipetted into the wells and FGF basic present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse FGF basic antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of FGF basic bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

  • Notes

FGF basic is a member of the fibroblast growth factor family. FGF family members bind heparin and possess broad mitogenic and angiogenic activities. FGF2 is involved in diverse biological processes, such as limb and nervous system development, wound healing, and tumor growth. FGF2 mRNA contains multiple polyadenylation sites, and is alternatively translated from AUG and non-AUG (CUG) initiation codons resulting in five different isoforms with distinct properties. The CUG-initiated isoforms are localized in the nucleus and are responsible for the intracrine effect, whereas, the AUG-initiated form is mostly cytosolic and is responsible for the paracrine and autocrine effects of this FGF.
