Overview

  • Product nameFGF basic protein (Active)See all FGF basic proteins and peptides ...
  • Protein descriptionRecombinant full length mature human FGF basic with His tag produced in the endosperm tissue of barley grain (Hordeum vulgare); amino acids 134-288 (156aa, 172aa with tag), 19.5 kDa. Swiss ID = P09038. Note: barley grain (Hordeum vulgare) exhibits up to 50 times less protease activity than E.coli or mammalian cells. Barley seed is void of any human or animal viral contaminants that could jeopardize your cell culture.
  • Expression hostBarley
  • Properties

  • Purity> 95 % by SDS-PAGE
  • Purification notesSterile filtered through a 0.2 µm filter. Endotoxin level is less than 0.005ng per µg (0.05EU/µg) as measured by turbidimetric kinetic assay. ab88270 carries no pyrogenic or pro-inflammatory contaminants, as assayed with monocyte activation test using Human 10-plex Cytokine Assay measuring IL-6, TNF-alpha and IL-1beta induction.
  • Biological activityBioactivity: The bioactivity is determined by dose-dependent effect of human FGF asic on proliferation of BaF3 cells transfected with FGF receptor. We recommend that the optimal concentration for each specific application be determined by an initial dose-response assay. ED50 determined from the BaF3 bioassay, is 1.59 ng/ml, corresponding to 0.62 x 106 U/mg.
  • FormLyophilised:It is recommended to reconstitute the lyophilized protein in sterile water to a concentration of no less than 100 µg/ml. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
  • Storage instructionsShipped at 4°C. Upon delivery aliquot and store at -20°C. Avoid repeated freeze / thaw cycles. This product is an active protein and may elicit a biological response in vivo, handle with caution.
  • Storage bufferPreservative: None
Constituents: 50mM Sodium chloride, 20mM Tris, pH 7.5

  • FunctionPlays an important role in the regulation of cell survival, cell division, angiogenesis, cell differentiation and cell migration. Functions as potent mitogen in vitro.
  • Tissue specificityExpressed in granulosa and cumulus cells. Expressed in hepatocellular carcinoma cells, but not in non-cancerous liver tissue.
  • Sequence similaritiesBelongs to the heparin-binding growth factors family.
  • Post-translational
    modifications
Phosphorylation at Tyr-215 regulates FGF2 unconventional secretion.
Several N-termini starting at positions 94, 125, 126, 132, 143 and 162 have been identified by direct sequencing.
  • Cellular localizationSecreted. Nucleus. Exported from cells by an endoplasmic reticulum (ER)/Golgi-independent mechanism. Unconventional secretion of FGF2 occurs by direct translocation across the plasma membrane. Binding of exogenous FGF2 to FGFR facilitates endocytosis followed by translocation of FGF2 across endosomal membrane into the cytosol. Nuclear import from the cytosol requires the classical nuclear import machinery, involving proteins KPNA1 and KPNB1, as well as CEP57.
