    Abcam's Human Fas Ligand ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of human Fas Ligand in serum, plasma, cell culture supernatants.

     

    This assay employs an antibody specific for human Fas Ligand coated on a 96-well plate. Standards and samples are pipetted into the wells and Fas Ligand present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-human Fas Ligand antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Fas Ligand bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

  • RelevanceFas / APO1 (CD95) is an important member of the tumour necrosis factor (TNF) superfamily involved in membrane-mediated apoptosis. Ligation of Fas by Fas Ligand or an anti Fas cross linking antibody, triggers activation of the caspase cascade. Functional impairment of the Fas / Fas Ligand system is associated with the development and progression of malignancies. Fas gene mutations have been suggested to have a role in testicular germ cell tumours. Tumour cells frequently exhibit de novo expression of Fas Ligand, which plays a significant role in local tissue destruction, metastatic spread, and immune escape of the tumor cells. The apoptosis of lymphocytes, which occurs in autoimmune diseases, is usually induced by the Fas/Fas Ligand system.

    We would suggest testing a range of dilutions to find a dilution within the standard curve. A good range is neat, 1:5, 1:10, 1:20. In the protocol the suggested dilution for normal serum/plasma: 2-10 fold, again this needs to be optimized.

    Unfortunately, the OD standard is never twice the same, therefore, it is so important to run a fresh standard curve for EVERY assay. The OD needs to be measured, it cannot be not calculated.
