Organizes filamentous actin into bundles with a minimum of 4.1:1 actin/fascin ratio. Plays a role in the organization of actin filament bundles and the formation of microspikes, membrane ruffles, and stress fibers. Important for the formation of a diverse set of cell protrusions, such as filopodia, and for cell motility and migration.

ab15110 staining human ovarian cancinoma tissue sections by IHC-P. Sections were formaldehyde fixed and subjected to heat mediated antigen retrieval in citric acid (pH 6) prior to blocking with 1% BSA for 10 minutes at RT.  The primary antibody was diluted 1/200 and incubated with the sample for 2 hours.

ab15110 staining gut tissue sections from mouse embyo (approx. day 15) by IHC-P.  Sections were formaldehyde fixed and subjected to het mediated antigen retrieval in citrate buffer (pH 6) prior to blocking in 1% BSA for 10 at RT.  The primary antibody was diluted 1/100 and incubated with the sample for 16 hours.  A biotinylated goat anti-mouse antibody was used as the secondary.

Flow Cytometry-Anti-Fascin antibody [SPM133](ab15110)

Overlay histogram showing HeLa cells stained with ab15110 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab151109, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells ) used under the same conditions. Acquisition of >5,000 events was performed.
