We are interested in using this antibody on both mouse and human IHC/paraffin embedded tissues. What antibody replacement would you suggest?



We are also interested in ordering some positive control sections/slides for testing this and other antibodies. I couldn’t find any ordering information on your site (what tissues you offer, cost, etc). Could you please send me this information? Thank you!

I suggest ab53962, anti-GAB1. This is the only GAB1 antibody in our catalogue that is validated for reactivity with both mouse and human samples.

Thank you for the quick response. We are confident that the secondary/tertiary antibodies are working, since we were also staining some other sections for different primaries and got good, specific staining with less than 5’ of DAB development. If you offer it, we would like a replacement that we could try on our sections. Do you also offer any positive-control sections that we could try as well? Thank you!

ANSWER:

 

A replacement will not be a problem, but we do not offer free control slides in addition to the replacement. We do have sections of a variety of paraffin-embedded tissues for sale, if you are interested.

Can you please tell me the species of your tissues? Antibody ab129374 may be a good choice, assuming your samples are human.

We have been trying this antibody using paraffin-embedded tissue sections. We have tried positive controls including spleen and heart, and only seen some nonspecific staining in nuclei. We have used citrate buffer antigen retrieval, blocking in BSA/casein/PBS and avidin/biotin, primary antibody 1:50 O/N at RT, secondary biotinylated antibody at 1:750, and tertiary streptavidin antibody at 1:1500. DAB was used to develop the stain. Washes were done 3x at each interface of antibody addition with PBST for 10 minutes.

I need a positive control. Do all astrocytoma stain positive? Should I expect 100%?

Hello! I received the reply below regarding staining of astrocytoma with the GAB1 antibody:

There are many variables during the fixation and embedding process that can affect tissue quality, thus not all astrocytoma cases will stain positive using this product. We routinely screen cases to determine the suitability of the tissue for IHC.

Therefore, as long as the tissue is suitable for IHC, all astrocytoma should stain positive for GAB1. Please let me know if you have any further questions.
