Our Abpromise guarantee covers the use of ab426 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
IHC-P: 1/50 - 1/500.
IHC-Fr: 1/50 - 1/500.
WB: Use a concentration of 1 µg/ml. Detects a band of approximately 74 kDa (predicted molecular weight: 67 kDa).
IHC-FoFr: Use at an assay dependent concentration.
GABA Transporter 1/GAT1 is a gamma-amino butyric acid (GABA) transporter. After its release from interneurons in the CNS, the major inhibitory neurotransmitter GABA is taken up by GABA transporters (GATs). The predominant neuronal GABA transporter GAT1 is localized in GABAergic axons and nerve terminals, where it is thought to influence GABAergic synaptic transmission by removing GABA from the synaptic cleft.
Cell Membrane
Western blot - GABA Transporter 1 / GAT 1 antibody (ab426)

All lanes : Anti-GABA Transporter 1 / GAT 1 antibody (ab426) at 1 µg/ml
Lane 1 : Rat Hippocampus Tissue Lysate
Lane 2 : Spinal Cord (Rat) Tissue Lysate
Lane 3 : Brain (Rat) Tissue Lysate
Lysates/proteins at 10 µg per lane.
Secondary
Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (HRP), pre-adsorbed at 1/3000 dilution
developed using the ECL technique
Performed under reducing conditions.
Predicted band size : 67 kDa
Observed band size : 74 kDa (why is the actual band size different from the predicted?)
Exposure time : 30 seconds
GABA Transporter 1/ GAT 1 contains a number of potential glycosylation sites (SwissProt) which may explain its migration at a higher molecular weight than predicted.
Immunohistochemistry (Frozen sections) - GABA Transporter 1 / GAT 1 antibody (ab426)

ab426 staining the GABA transporter 1 in Mouse retinal tissue sections by Immunohistochemistry (IHC-Fr - frozen sections). Tissue was fixed with paraformaldehyde, and blocked with 5% serum for 1 hour at 20°C. Samples were incubated with primary antibody (1/2000 in PBST) for 12 hours at 4°C. An Alexa Fluor®488-conjugated Donkey anti-rabbit polyclonal (1/1000) was used as the secondary antibody.
