Applications

Show applications key

Our Abpromise guarantee covers the use of ab32858 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application notes

IHC-P: Use at a dilution of 1/50-1/100 for 1 hr.
WB: Use at a concentration of 1 µg/ml. Detects a band of approximately 51 kDa (predicted molecular weight: 48 kDa).

Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Target

Function

Transcriptional activator which binds to the enhancer of the T-cell receptor alpha and delta genes. Binds to the consensus sequence 5'-AGATAG-3'.

Tissue specificity

T-cells and endothelial cells.

Involvement in disease

Defects in GATA3 are the cause of hypoparathyroidism with sensorineural deafness and renal dysplasia (HDR) [MIM:146255]; also known as Barakat syndrome.

Sequence similarities

Contains 2 GATA-type zinc fingers.

Cellular localization

Nucleus.

Secondary
Goat polyclonal to Rabbit IgG - H&L - Pre-Adsorbed (HRP) at 1/3000 dilution

Performed under reducing conditions.

Predicted band size : 48 kDa
Observed band size : 51 kDa (why is the actual band size different from the predicted?)
Additional bands at : 42 kDa. We are unsure as to the identity of these extra bands.

Exposure time : 4 minutes

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - GATA3 antibody (ab32858)

ab32858 staining GATA3 in human thymus tissue sections by immunohistochemistry (Formalin/PFA-fixed paraffin embedded sections). Tissue underwent fixation in formaldehyde, heat mediated antigen retrieval in citrate buffer pH6.0 and blocking for 15 minutes at 20°C (5 minutes for peroxidase block and 10 minutes for protein block). The primary antibody was diluted 1/50 and incubated with sample for 45 minutes at 20°C. A HRP-conjugated goat polyclonal to rabbit IgG was used undiluted as secondary.

This antibody is part of an abreview submitted by Antibody Solutions Ltd.
