Mouse monoclonal to GFP
Assayed by ELISA for direct binding of antigen recognizes wild type and recombinant GFP. Well suited to titrate GFP in solution using either form of the antibody as the capture or detection antibodies. Shows no reactivity against RFP.
WB, IHC-Fr, Sandwich ELISA, ICC/IF, IPmore details
The immunogen is a Green Fluorescent Protein (GFP) fusion protein corresponding to the full length amino acid sequence (246aa) derived from the jellyfish Aequorea victoria.

IgG fraction
This product was prepared from tissue culture supernatant by gel filtration and ion exchange chromatography. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Mouse Serum and purified and partially purified Green Fluorescent Protein (Aequorea victoria). Reactivity is observed against wild type, recombinant and enhanced forms of GFP.
Monoclonal
IgG2a
Our Abpromise guarantee covers the use of ab1218 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
WB: 1/500 - 1/2000.((See Abreviews). Recognises the immunogen under native conditions and reduced/denatured conditions.)
IHC-Fr: 1/500(Works also with eYFP.)
sELISA: Use at an assay dependent dilution. Can be paired for Sandwich ELISA with Goat polyclonal to GFP (Biotin) (ab6658).
ICC/IF: Use at an assay dependent concentration.
IP: Use at an assay dependent dilution. (PubMed: 16135826)
Green fluorescence protein (GFP) is a 27 kDa protein derived from the jellyfish Aequorea victoria, which emits green light (emission peak at a wavelength of 509 nm) when excited by blue light (excitation peak at a wavelength of 395 nm). GFP has become an invaluable tool in cell biology research, since its intrinsic fluorescence can be visualized in living cells. GFP fluorescence is stable under fixation conditions and suitable for a variety of applications. GFP has been widely used as a reporter for gene expression, enabling researchers to visualize and localize GFP-tagged proteins within living cells without the need for chemical staining. Other applications of GFP include assessment of protein protein interactions through the yeast two hybrid system and measurement of distance between proteins through fluorescence energy transfer (FRET) protocols. GFP technology has considerably contributed to a greater understanding of cellular physiology. YFP differs from GFP due to a mutation at T203Y; antibodies raised against full-length GFP should also detect YFP and other variants.
Immunoprecipitation - GFP antibody (ab1218)

Immunoprecipitation using ab1218 at 0.01µl/µl HEK (human embryonic kidney) 293 whole cell lysate incubated with protein G matrix for 1 hour at 4°C.
Total protein input was 3,000,000 cells.
Lane 1: GFP-RhoA wt
Lane 2: GFP-Rac1 wt
Lane 3: GFP-empty
The western blot antibody was ab290 (undiluted).

Immunocytochemistry/ Immunofluorescence - GFP antibody (ab1218)

ab1218 staining GFP in human prostate epithelial primary cells by Immunocytochemistry/ Immunofluorescence. Cells (ARPE-19 cells transfected with mouse Rab27A-GFP) were fixed with paraformaldehyde, permeabilized with 0.5% Triton x100 and blocking with 5% serum was performed for 20 minutes at 25°C. Samples were incubated with primary antibody (1/500: in 1% goat serum, 0.1%TX100, 1XPBS) for 16 hours at 4°C. An Alexa Fluor®546-conjugated goat polyclonal to mouse IgG was used as secondary antibody at 1/500 dilution. DAPI was used to stain the cell nuclei (blue) and mRab27A-GFP with ab1218 (red).
