Our Abpromise guarantee covers the use of ab84403 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
WB: 1/200 - 1/500.Predicted molecular weight: 182 kDa.
ELISA: Use at an assay dependent dilution.
IHC-P: Use a concentration of 1 µg/ml
Plays a role in RNA-mediated gene silencing by both micro-RNAs (miRNAs) and short interfering RNAs (siRNAs). Required for miRNA-dependent repression of translation and for siRNA-dependent endonucleolytic cleavage of complementary mRNAs by argonaute family proteins.
Ubiquitous.
Belongs to the GW182 family.
Contains 1 RRM (RNA recognition motif) domain.
Cytoplasm > P-body. Mammalian P-bodies are also known as GW bodies.

Western blot - GW182 antibody (ab84403)

Anti-GW182 antibody (ab84403) at 1/200 dilution + human fetal brain lysates
Predicted band size : 182 kDa
Observed band size : 182 kDa
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-GW182 antibody (ab84403)

IHC image of ab84403 staining in Breast Cancer formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab84403, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
