 I am interested to know if this product (Ab11081) has been used with any other anti-gelsolin antibodies as part of a sandwich ELISA?   Kind regards,

Thank you for your inquiry and please excuse the delayed answer.

I was in contact with the source of this antibody and did a literature search, but I could not find any other anti-gelsolin antibodies that were used with ab11081 as part of a sandwich ELISA.

We have two other polyclonal antibodies against gelsolin that are tested in ELISA so far.

Can you tell me the range of immunogen used to generate mouse monoclonal [GS-2C4] anti human gelsolin antibody (ab11081)?

Thank you for contacting us. The immunogen is the full-length human gelsolin, and the epitope that it binds to is located somewhere on the carboxyl-terminal two-thirds. The following reference describes production of monoclonal antibodies against human plasma gelsolin, including this clone, 2C4.

From page 1476:

"This (47 kD) peptide, and a 45-kD peptide with similar actin-binding properties reacted with the monoclonal antibody designated 2C4 (Fig. 2 c). These two peptides are therefore related and contain the same actin-binding domain which is shown in Kwiatkowski et al. (1985) to be derived from the carboxyl-terminal two-thirds of plasma gelsolin."

What is the shelf life of gelsolin Ab? We intend to buy in bulk to last us for 2 years but is concern that the Ab might not work if stored too long.

I would like to known if this antibody is suitable to use in human platelets or if it can only be used in plasma. Further does it appear in reducing conditions as a 86 KDa band?

Ab11081 reacts specifically with human plasma and cytoplasmic gelsolin in immunoblotting. The antibody is directed against an epitope located on the 47 kD peptide derived from a chymotryptic cleavage of gelsolin. This peptide contains the carboxy terminal actin binding site for gelsolin. Gelsolin has been shown to exist in at least two variant forms, cytoplasmic gelsolin (from rabbit macrophages and human platelets) and plasma gelsolin (from human or bovine plasma). Human plasma and rabbit macrophage gelsolins differ by the presence of a 25 amino acid residue extension at the NH2 terminus in the human plasma gelsolin which appears to account for the difference in relative molecular weights. I think this should work on human platelets but you may have to work on the titration for using it. We have seen two doublet bands at 97 kDa and it's possible that you will see a lower band under reduced conditions.

For ab2500, you should see a band at approximately 210 kD.

To whom it may concern, I'm tryng to find out if gelsolin is involved in the rearrangement of actin cytoskeleton that I have detected in rabbit skeletal muscle. I'll do so by immuno fluorescence. I would like to know if this monoclonal anti-gelsolin antibody will recognize the protein when bound to actin, since the epitop that is raised against contains the actin binding domain I'm afraid it may be blocked when actin is bound to the domain?

I can confirm that ab11081 will detect both cytoplasmic gelsolin and actin-bound gelsolin.
