I was studying a lot Gli2 and was trying to choose Gli2 antibody which would be correct to use to detect active form of Gli2 .

As you may know human Gli2 has N-terminus region (repressor), DNA binding Zn finger region [437-589 aa] (this info is from Ensemble and from UniProtKB) and C-terminus region (activator) [590-1586]. Despite of the fact that Gli2 is presented in 5 isoforms, DNA biding region is conservative

1586 aa – 168 kDa (full length) 1258 aa – 133 kDa (1-328: Missing) 1241 aa – 132 kDa (1-328; Missing; 394-410: Missing) 829 aa – 88 kDa (1-328: Missing; 1149-1157: VSSGTVDAL → ASATWLSGT; 1158-1586: Missing) 812 aa – 86.5 kDa (1-328: Missing; 394-410: Missing; 1149-1157: VSSGTVDAL → ASATWLSGT; 1158-1586: Missing)

In addition, post-translational modifications (such as acetylation which is important for Gli2 activity etc.) and mutations might also change presented above Gli2 MW in different cancers and/or cell types.

Your anti-Gli2 antibody (ab26056) is ChIP grade. How it is possible if this antibody was generated using as immunogen a synthetic peptide derived from a region between residues 272 - 321 (which is within N-terminus, and for Gli2 activation, this region should be degraded)?

After review of the materials for this product I have found that the immunogen sequence location provided was indeed incorrect. The immunogen was created from the area aa 600-649 ofhuman Gli2 (SwissProt.P10070). I have requested that this information be changed on the datasheet as well.
