- Specificityab85715 recognizes Glucose Transporter GLUT2 expression on hGlut2-transfected NS0 cells but not on control transfectants. Detects Glut2 on the surface of the Human intestinal cell line Caco2 in flow cytometry and on fixed cells in immunocytochemistry tests. Based on flow cytometric tests on transfected cells, this antibody has no cross-reactivity with hGlut1 or hGlut3.

Target
- FunctionFacilitative glucose transporter. This isoform likely mediates the bidirectional transfer of glucose across the plasma membrane of hepatocytes and is responsible for uptake of glucose by the beta cells; may comprise part of the glucose-sensing mechanism of the beta cell. May also participate with the Na(+)/glucose cotransporter in the transcellular transport of glucose in the small intestine and kidney.

Overlay histogram showing HepG2 cells stained with ab85715 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab85715, 0.5µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG2a [ICIGG2A] (ab91361, 1µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in HepG2 cells fixed with 80% methanol (5 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.

Glucose Transporter GLUT2 was stained using ab85715 (green), at 10 mg/ml. An AlexaFluor®647-conjugated donkey anti-mouse IgG was used as the secondary antibody.
