H6PD (hexose-6-phosphate dehydrogenase) oxidizes glucose-6-phosphate and glucose, as well as other hexose-6-phosphates. Defects in H6PD are a cause of cortisone reductase deficiency, in which the activation of cortisone to cortisol does not occur, resulting in a phenotype resembling polycystic ovary syndrome (PCOS). H6PD interacts with 11 beta-hydroxysteroid dehydrogenase type I, and both proteins may play important roles in the pathogenesis of obesity and metabolic syndrome.

Predicted band size : 88 kDa Observed band size : 94 kDa (why is the actual band size different from the predicted?)
Exposure time : 2 minutes
Human GDH/6PGL endoplasmic bifunctional protein precursor (H6PD) has a predicted molecular weight of 88 kDa, however it is glycosylated and phosphorylated which may explain the bands appearing at a higher molecular weight (94 kDa)(SwissProt data).

Immunocytochemistry/ Immunofluorescence - H6PD antibody (ab72183)
ICC/IF image of ab72183 stained HeLa cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal Goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab72183, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 Goat anti-Rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM. This antibody also gave a positive result in 4% PFA fixed (10 min) HepG2, and MCF-7 cells at 5µg/ml.
