Overview

  • Product nameHEC1 protein (Tagged-His Tag)See all HEC1 proteins and peptides ...
  • Protein descriptionRecombinant fragment, corresponding to amino acids 350-642 (293 amino acids) of Human HEC1 with N terminal His tag; MWt 37 kDa (UniProt O14777).
  • Expression hostE. coli
  • Properties

  • Purification notesPurity level >85%
This protein was expressed as an N-terminal His-tag fusion protein using Escherichia coli, and purified using Immobilized Metal Ion Affinity Chromatography. In some cases, smaller protein fragments may be present in addition to the intended expression product as a result of premature termination during translation in E. coli and subsequent co-purification via the His-tag. In some cases purified proteins run at a molecular weight different to the theoretically calculated molecular weight. This may be as a result of unequally distributed charges in the amino acid sequence. Alternatively, dimerisation of the expression product can occur under oxygen limitation during expression/cultivation.

  • FunctionActs as a component of the essential kinetochore-associated NDC80 complex, which is required for chromosome segregation and spindle checkpoint activity. Required for kinetochore integrity and the organization of stable microtubule binding sites in the outer plate of the kinetochore.
  • Sequence similaritiesBelongs to the NDC80/HEC1 family.
  • Developmental stageExpression peaks in mitosis.
  • Post-translational
    modifications
Phosphorylation begins in S phase of the cell cycle and peaks in mitosis. Phosphorylated by NEK2. May also be phosphorylated by AURKA and AURKB.
  • Cellular localizationNucleus. Chromosome > centromere > kinetochore. Localizes to kinetochores from late prophase to anaphase. Localizes specifically to the outer plate of the kinetochore.
