Recommended dilutions IHC-P: 1/1000. Antigen retrieval is not essential but may optimise staining. See Zong et al 2001 reference (or protocol at http://www.ihcworld.com/_protocols/antibody_protocols/hif_1alpha_novus.htm). Optimal antigen retrieval method: Briefly, pre-heat steamer or water bath with staining dish containing 10 mM citrate buffer pH 6.0 until temperature reaches 95-100°C. Immerse slides in the staining dish and place the lid loosely on the staining dish. Incubate for 45 minutes and turn off the steamer or water bath. Place the staining dish at room temperature and allow the slides to cool for 20 minutes. ICC/IF: See recommended protocol below. IP: Use at an assay dependent dilution. WB: Use at an assay dependent dilution.
Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Oxygen availability plays a major role in the regulation of expression of many different genes including erythropoietin, nitric oxide synthase (NOS), heme oxygenase 1 (HO-1), glucose transporters and vascular growth factors necessary for the maintenance of homeostasis in hypoxic conditions. Hypoxia-inducible factor 1 alpha (HIF1 alpha) has been identified as a bHLH-PAS family member that is instrumental in the oxygen-dependent regulation of these genes. HIF1 alpha rapidly accumulates in the nucleus upon exposure to hypoxic conditions where it heterodimerises with the aryl hydrocarbon nuclear receptor translocator ARNT (also referred to as HIF1 beta). Heterodimerisation promotes efficient DNA binding.
